The anther is the site where male gametophytes (pollen grains) develop. A typical anther is bilobed (dithecous) and, with two theca per lobe, is tetrasporangiate โ it contains four microsporangia that mature into pollen sacs.
Microsporangium structure
In transverse section a microsporangium has four wall layers: the epidermis, endothecium, one to three middle layers, and the innermost tapetum. The outer three layers protect and help dehiscence (the endothecium aids splitting), while the tapetum nourishes the developing pollen โ its cells have dense cytoplasm and often more than one nucleus. The centre holds the sporogenous tissue.
Microsporogenesis and the pollen grain
Each cell of the sporogenous tissue is a microspore mother cell (MMC). It undergoes meiosis to form a microspore tetrad; the microspores separate and develop into pollen grains โ this is microsporogenesis. The pollen grain (male gametophyte) is 25โ50 ยตm across with a two-layered wall: a hard outer exine made of sporopollenin (the most resistant known organic material, unaffected by high temperature, acids or alkali) bearing germ pores, and an inner celluloseโpectin intine. At maturity most pollen has two cells: a large vegetative cell with food reserves and a small generative cell that divides to form two male gametes. About 60% of angiosperms shed pollen at the 2-celled stage; the rest shed at the 3-celled stage.

Microsporangium and pollen grain (male gametophyte)
Inside every anther, thousands of dust-like pollen grains are being built โ each one a tiny, wall-armoured, two-to-three-celled male plant. This is one of biology's most compact forms of life.
Core idea: The pollen grain is the male gametophyte of angiosperms. It develops inside the microsporangium of the anther through microsporogenesis followed by mitotic division of the microspore.
Anther and microsporangium
Structure
A typical anther is dithecous (two lobes) and tetrasporangiate โ four microsporangia arranged at the corners. Each microsporangium is surrounded, outside-in, by four wall layers:
- Epidermis โ protective.
- Endothecium โ has fibrous thickenings; helps in dehiscence (splitting to release pollen).
- Middle layers โ 1โ3 layers, ephemeral.
- Tapetum โ the innermost, densely cytoplasmic nutritive layer that nourishes developing pollen and provides sporopollenin and the exine pattern.
At the centre lies the sporogenous tissue.
Microsporogenesis
The process of forming microspores from a pollen mother cell (PMC) by meiosis:
PMC (2n) โ meiosis โ a microspore tetrad (four haploid microspores) โ the tetrad separates into individual microspores.

Wall
- Exine โ the hard outer wall made of sporopollenin, the most resistant organic material known (unaffected by high temperature, acids, alkalis, enzymes). It bears germ pores. Its durability is why pollen fossilises well.
- Intine โ a thin, continuous inner wall of cellulose and pectin.
Cells
The microspore divides mitotically to form:
- A large vegetative cell โ abundant food reserve, irregular nucleus; forms the pollen tube.
- A small generative cell โ spindle-shaped, floats in the vegetative cytoplasm; divides to form two male gametes.
In ~60% of angiosperms pollen is shed at the 2-celled stage; in the rest the generative cell divides before shedding, giving a 3-celled pollen grain.
Worked example
Q: A pollen grain is shed at the 3-celled stage. Name the cells present.
A: One vegetative cell and two male gametes (the generative cell has already divided). Such pollen is 'ready to fertilise' on landing.
Real-world / exam application
Pollen viability varies โ rice/wheat pollen lives minutes, while Rosaceae/Leguminosae pollen survives months. Pollen banks store pollen in liquid nitrogen (cryopreservation) for crop breeding, just as we store seeds. Pollen allergies (e.g. Parthenium) and 'pollen tablets/syrups' as food supplements are also asked.

Do not confuse the tapetum (nutritive wall layer) with the sporogenous tissue (the cells that undergo meiosis). Also, a microspore is not the same as a pollen grain until its wall matures โ but a single microspore becomes a single pollen grain (no fusion).

Problem: Explain how a pollen grain (the male gametophyte) develops from the microsporangium, and describe the structure of a mature pollen grain at the time of shedding.
Solution:
Step 1 โ Start at the microsporangium. Inside the anther, the microsporangium contains sporogenous tissue whose cells act as pollen mother cells (microspore mother cells).
Step 2 โ Describe microsporogenesis. Each diploid pollen mother cell undergoes meiosis to produce a tetrad of four haploid microspores. Each microspore matures into a pollen grain.
Step 3 โ Describe the pollen wall. The mature pollen grain has a two-layered wall: a hard, resistant outer exine (made of sporopollenin, often sculptured, with germ pores) and an inner cellulose intine.
Step 4 โ Describe the contents at shedding. In most species the pollen is shed at the 2-celled stage: a large vegetative cell (with abundant food reserve, forms the pollen tube) and a smaller generative cell (which later divides to form two male gametes).
Answer: Meiosis of pollen mother cells in the microsporangium gives haploid microspores that mature into pollen grains; a mature pollen grain has an exine and intine wall and is usually 2-celled (a vegetative and a generative cell).
